- COMPOSITION:
Phenobarbital 100 mg/1 ml; 200 mg/2 ml in one ampoule; 20 mg/in
5 ml syrup and 15 mg and 100 mg in one tablet.
ACTION:
Phenobarbital suppresses CNS and ahs a sedative hypnotic and
anticonvulsive action, depending on the dose. Compared with other
barbiturates, it inhibits more strongly the excitation of the motor
centers of the brain, hence it is widely used in the treatment of
epilepsy, chorea and spastic paralysis. As a sedative (usually in
combinations), it is administered in low doses for the treatment of
the initial stages of hypertonic disease and vascular spasms
(migraine, angina pectoris). As a drug with acid character it has
higher concentrations in the extracellular fluids. With the increase
of pH values in the medium, the percentage share of the ionized part
of phenobarbital abruptly increases. Its protein content is the
highest in adults and the lowest in the fetus. Phenobarbital is one
of the active enzyme inductors. The coefficient of its liver
elimination (main pathway) is rather low; from 0.1 to 0.3. The
analysis of urine leads to its intensive ionization and to enhanced
excretion.
INDICATIONS:
Epilepsy, chorea, sleep disturbances. In small doses in hypertonic
disease. It has a good effect on vascular spasms (migraine, angina
pectoris).
CONTRAINDICATIONS:
Diseases of liver and kidneys, high temperature, allergy to
barbiturates, the first three months of pregnancy.
SIDE EFFECTS:
The high doses can cause a suppression of respiration, vascular
collapse, pulse deceleration; temperature decrease, reduction of
diuresis; lassitude, apathy, ataxia, memory impairment, psychic
disturbances, allergic reactions. |
PRECAUTIONS:
To be cautiously prescribed to drivers or professions requiring fast
psychic and motor reactions. Do not prescribe to breast-feeding
mothers, the drug passes into the mother's milk.
DRUG INTERACTION:
When administered with phenobarbital, the metabolism of certain
drugs is accelerated, their effect becoming weaker and shortens.
More than 60 chemical substances react in this way, among them
anticoagulants, digitoxin, corticosteroids, many of the non-steroid
antiphlogistic agents, theophyllin (phenomenon of
"induction" of enzymes).
DOSAGE AND ADMINISTRATION:
In insomnia it is orally prescribed as a hypnotic drug, if the
insomnia is associated with anxiety and excitation - adults:
0.1-0.2, a single intake in the evenings; children: 0.005-0.075 per
intake, depending on age. In epilepsy it is administered twice daily
- 0.05 with gradual increase of the dose, but not more than 0.6 g
per 24 h; as a spasmolytic and sedative - 0.01-0.03 g, 2-3 times
daily.
MEDICINAL FORM AND PACKAGE:
Packages with 10 and 50 ampoules of 100 mg/1 ml and 200 mg/2 ml.
Syrup in vials of 120 ml.
Packages with 30 tablets of 100 mg.
STORAGE:
At moderate temperature (15-30 C) in a place protected from light.
EXPIRY:
Ampoules and syrup - 3 (three) years.
Tablets - 5 (five) years. |
